Work= Western + cast gels
Okies, that basically roughly sums what ilve been doing all these while. Okay, there are other stuff like mini preps and cell harvesting and stuff but casting gel and this technique called western blot is what i always do , without fail, rain or shine. it seems like my life revolve around them.
Ok, so it is logical to say that after casting almost a hundred gels and developing them for Western blots, i should be quite good at it right? but NO! recently my gels and blots have been giving me lots of problems!!!
I dont know why funny stuff always happens to me. Last time was the eppendorf tube cracking for no apparent reason, resulting in me losing my sample. Now, new stuff happens! I keep casting weird gels which have funny shapes! and my blots are terrible! one minute the signal is too weak and when i used a stronger detector which shd give me a stronger signal, the signal totally dissapeared!!! WHY WHY WHYYEEE? this is so odd! and today i tried casting the gel over and over again and it didnt work! i did it three times today! It isnt so bad when you cast and get your prettu gel but today i casted 3 times and because it wasnt good so i have to throw away and cast! so it means i have to disassemble the apparatus, scrap away the gel, wash the plates with soap and dry it, clean with ethanol, and assemble it and lastly make the solution again!Not to mention to pour it.
Work is getting quite monotonous, especially when I dont know whats the rationale behind all these things i;m doing. I want to ask my metor! But he's jus too busy with his work. Sometimes i feel that he is guqite unlucky to get me as his student. Poor guy! i'm so sorry.
But it's really amazing how i got the job. :) I just sent them an email and they took me! yay! without any interviews! I guess it's God's grace and i really need to be thankful for that, Cause i know people who applied but wasnt given the job coz they dont need people already.
So, overall, i;m still happy with my job. :) and my friend say i look v happy there, haha...maybe i am! :) but i really ho[e i can find out what's wrong with my gel and blots. :( haha
